由大丽轮枝菌(Verticillium dahliae)侵染所引起的棉花黄萎病,每年给我国农业生产带来巨大的经济损失.同时,棉花与大丽轮枝菌互作过程中发生的免疫反应变化及其机理尚不明确,缺乏棉花免疫反应研究的指示基因.本研究根据已报道的植物病程相关基因(pathogenesis-related gene,PR基因)序列,搜索陆地棉(Gossypium hirsutum)基因组数据库,获得14个潜在的PR基因家族(GhPR).为了明确GhPR在植物免疫反应过程中的表达情况,分别利用两种已报道的病原相关分子模式(PAMP)肽段flg22和nlp20,以及3个来自大丽轮枝菌、与nlp20(nlp20~(Pp))同源的肽段nlp20~(Vd2),nlp23~(Vd3),以及nlp23~(Vd4),对无菌棉花根部进行处理,检测GhPR基因在不同处理时间段的表达情况.结果显示,在flg22,nlp20~(Pp),nlp20~(Vd2),nlp23~(Vd3)和nlp23~(Vd4)处理下,GhPR基因普遍上调表达,且在处理后3 h达到最高值;在处理后12 h内,nlp20~(Vd2)比其他肽段更为有效地诱导GhPR基因上调表达;尽管nlp23~(Vd3)和nlp23~(Vd4)所在的全长蛋白VdNLP3和VdNLP4不能诱导棉花叶片的坏死反应,但是这两个肽段却能够引起GhPR基因的上调表达.因此,本研究为棉花-大丽轮枝菌的互作的研究提供了免疫反应指示基因GhPR,以及来自大丽轮枝菌的免疫原性肽段nlp20~(Vd2),nlp23~(Vd3),以及nlp23~(Vd4).

Although the full-length proteins VdNLP3 and VdNLP4 of nlp23 ~ (Vd3) and nlp23 ~ (Vd4) did not induce the necrosis reaction of cotton leaves, both of them could up-regulate the expression of GhPR gene . Therefore, the present study provides an immune response indicator gene GhPR for the interaction between cotton and Dahliae, and the immunogenic peptide nlp20 ~ (Vd2), nlp23 ~ (Vd3) , And nlp23 ~ (Vd4).
