目的 神经生长因子 (nerve growth factor, NGF)和脑源性神经营养因子( brain- derived neurotrophic factor, BDNF)是具有感觉和运动神经元营养活性的因子。为提高雪旺细胞 (Schwann cell, SC)分泌 NGF和 BDNF的能力,将 NGF和 BDNF基因转入 SC进行探讨。方法 用消化后的组织块法和差速离心法分离、纯化雪旺细胞。用脂质体转染法将 NGF和 BDNF基因导入雪旺细胞, ELISA双抗体夹心法检测 NGF和 BDNF的表达。结果 经免疫组化 S- 100法鉴定 SC纯度达 95%以上。 ELISA双抗体夹心法测定转染后不同时期的 NGF和 BDNF的浓度, 2周后表达增加, 4周后明显增加,差异有非常显著性意义 (P
Objective Nerve growth factor (NGF) and brain-derived neurotrophic factor (BDNF) are the factors that influence the nutritional activity of sensory and motor neurons. To enhance the ability of Schwann cells to secrete NGF and BDNF, NGF and BDNF genes were transferred into SC cells. Methods The digested tissue block method and differential centrifugation method were used to isolate and purify Schwann cells. NGF and BDNF genes were introduced into Schwann cells by lipofection method, and the expression of NGF and BDNF was detected by ELISA double antibody sandwich method. Results The purity of SC was identified as 95% by immunohistochemistry. The concentration of NGF and BDNF in different periods after transfection was detected by ELISA double antibody sandwich method.
