目的探讨T2DM对OLETF大鼠不同器官组织胶原蛋白含量的影响。方法选取造模成功的T2DM OLETF大鼠(DM组)16只和同系LETO鼠(NC组)12只,分别采集皮肤、肝脏、股骨组织,进行HE、Masson染色,免疫组织化学法及高效液相色谱法测定胶原蛋白含量。结果与NC组相比,DM组皮肤、股骨组织胶原纤维减少,肝脏胶原纤维增多。DM组每克皮肤中胶原蛋白含量[(149.9±18.4)vs(266.0±54.8)mg/g]和股骨组织胶原含量[(185.60±19.38)vs(243.27±22.19)mg/g]均低于NC组(P<0.05或P<0.01);而肝脏胶原蛋白含量高于NC组[(134.54±18.78)vs(64.52±21.51)mg/g,P<0.01]。结论 T2DM对不同组织胶原蛋白含量影响不同,皮肤、股骨胶原蛋白含量减少,肝脏胶原蛋白含量增加。
Objective To investigate the effect of T2DM on the collagen content in different organs of OLETF rats. Methods Twenty - two T2DM OLETF rats (DM group) and 12 LETO mice (NC group) were enrolled in this study. Skin, liver and femur tissues were collected and stained with HE, Masson staining, immunohistochemistry and high performance liquid chromatography Chromatographic determination of collagen content. Results Compared with NC group, collagen fibers in the skin and femur of DM group decreased and hepatic collagen fibers increased. Collagen content per gram of skin [(149.9 ± 18.4) vs (266.0 ± 54.8) mg / g] and femoral collagen content [(185.60 ± 19.38) vs (243.27 ± 22.19) mg / g] (P <0.05 or P <0.01), while the content of collagen in liver was higher than that in NC group [(134.54 ± 18.78) vs (64.52 ± 21.51) mg / g, P <0.01]. Conclusions T2DM has different effects on the collagen content of different tissues, the content of collagen in the skin and femur decreased, and the content of collagen in the liver increased.
