采用通用引物及聚合酶链反应(PCR)法,从分泌抗胃癌单克隆抗体的杂交瘤细胞3H11总RNA中分别扩增轻重链可变区基因并克隆入相应载体。序列分析表明SH11VL属VkIV亚群,J源于JK4。3H11VH属VHIIB亚群,J及D区分别来自JH4及DSP2。
Using universal primers and polymerase chain reaction (PCR), the light and heavy chain variable region genes were amplified from hybridoma 3H11 total RNA secreting anti-gastric cancer monoclonal antibody and cloned into the corresponding vector. Sequence analysis showed that SH11VL belongs to VkIV subgroup and J originates from JK4. 3H11VH belongs to VHIIB subpopulation and J and D regions come from JH4 and DSP2, respectively.
