[目的]观察白藜芦醇对脂多糖刺激小鼠RAW264.7细胞的NF-κB核转位影响。[方法]体外培养小鼠RAW264.7细胞,脂多糖刺激小鼠RAW264.7细胞,用不同浓度白藜芦醇进行干预,免疫荧光法结合激光共聚焦显微镜检测NF-κB核转位情况。[结果]共聚焦显微镜下观察到正常组细胞NF-κB荧光标记主要分布于胞浆区,核区很少,LPS刺激后NF-κB荧光标记浓集于核区,胞浆区少见;白藜芦醇预处理后再给予LPS刺激的荧光标记主要位于胞浆区,以100、50μmol.L-1组较明显。[结论]白藜芦醇能抑制脂多糖刺激下小鼠RAW264.7细胞的NF-κB核转位。
[Objective] To investigate the effect of resveratrol on nuclear translocation of NF-κB induced by lipopolysaccharide in mouse RAW264.7 cells. [Method] Mouse RAW264.7 cells were cultured in vitro, and lipopolysaccharide was used to stimulate mouse RAW264.7 cells. Different concentrations of resveratrol were used for intervention. The nuclear translocation of NF-κB was detected by immunofluorescence and laser confocal microscopy. [Results] The results of confocal microscopy showed that the NF-κB fluorescent markers of normal cells were mainly distributed in the cytoplasm, with few nuclear regions. The NF-κB fluorescent markers were concentrated in the nucleus after LPS stimulation. Fluorescent labeling of LPS stimulated with resveratrol was mainly located in the cytoplasm, especially in the groups of 100 and 50 μmol.L-1. [Conclusion] Resveratrol can inhibit NF-κB nuclear translocation in mouse RAW264.7 cells stimulated by lipopolysaccharide.
