目的探讨白藜芦醇对人胃癌MGC-803细胞株细胞周期影响及其机制。方法采用四甲基偶氮噻唑蓝(MTT)法和集落形成试验检测白藜芦醇对胃癌细胞的增殖抑制作用,流式细胞术检测细胞周期的改变,逆转录-聚合酶链式反应(RT-PCR)检测细胞凋亡抑制基因-2(bcl-2)、增殖基因c-myc的mRNA表达水平。结果白藜芦醇可抑制胃癌细胞增殖和集落形成能力,低、高剂量组的抑制率分别为6.9%,17.6%和27.98%,63.39%,差异均有统计学意义(P<0.05);白藜芦醇能使胃癌细胞周期阻滞于G0/G1期,明显下调胃癌细胞bcl-2、c-myc基因mRNA表达水平(P<0.05);用药24 h后,白藜芦醇高剂量组bcl-2和c-myc基因相对表达量分别为(0.54±0.04),(0.58±0.05),与空白对照组(0.91±0.05),(0.85±0.03)比较,差异均有统计学意义(P<0.05)。结论白藜芦醇可抑制胃癌MGC-803细胞增殖并使其细胞周期阻滞于G0/G1期,可能与下调bcl-2、c-myc基因表达有关。
Objective To investigate the effect and mechanism of resveratrol on cell cycle of human gastric cancer cell line MGC-803. Methods MTT assay and colony formation assay were used to detect the effects of resveratrol on the proliferation of gastric cancer cells. Flow cytometry was used to detect the changes of cell cycle, RT-PCR (RT-PCR) -PCR) were used to detect the mRNA expression levels of apoptosis-inhibiting gene-2 (bcl-2) and proliferation gene c-myc. Results Resveratrol could inhibit the proliferation and colony forming ability of gastric cancer cells. The inhibitory rates of resveratrol were 6.9%, 17.6% and 27.98%, 63.39%, respectively, with significant difference (P <0.05) Resveratrol could block the cell cycle of gastric cancer cells in G0 / G1 phase and significantly decrease the mRNA expression of bcl-2 and c-myc in gastric cancer cells (P <0.05). After 24 h, resveratrol high-dose group (0.54 ± 0.04) and (0.58 ± 0.05), respectively, which were significantly different from those in control group (0.91 ± 0.05) and (0.85 ± 0.03), respectively (P < 0.05). Conclusion Resveratrol can inhibit the proliferation of gastric cancer cell line MGC-803 and arrest its cell cycle in G0 / G1 phase, which may be related to down-regulation of bcl-2 and c-myc gene expression.
