目的:研究眼镜蛇毒直接溶解因子(DLF)对人肝癌细胞株H7402产生一氧化氮(NO)和脂质过氧化物(LPO)的影响。结果:DLF对体外培养的H7402细胞有杀伤作用,IC50为247mg·L-1,DLF与H7402细胞的生长抑制率呈量效关系。DLF可介导H7402细胞产生大量NO2,NO2量与DLF的剂量呈量效关系。在一定剂量范围内NO2量与H7402的生长抑制率呈显著正相关。用570型粘附式细胞仪测定单个细胞内LPO,发现DLF可使H7402细胞内LPO产生增加。结论:DLF可杀伤H7402细胞,其部分机制可能是通过诱导肿瘤细胞产生NO和LPO。
OBJECTIVE: To investigate the effects of direct lytic factor (DLF) from cobra venom on the production of nitric oxide (NO) and lipid peroxides (LPO) in human hepatoma cell line H7402. RESULTS: DLF had killing effect on H7402 cells cultured in vitro with IC50 of 2.47 mg·L-1. The growth inhibitory rate of DLF and H7402 cells was dose-effect relationship. DLF can mediate a large amount of NO 2 in H7402 cells. The dose of NO 2 and DLF dose-effect relationship. There was a significant positive correlation between the amount of NO 2 and the growth inhibition rate of H7402 within a certain dose range. Using a 570-type adherent cytometer to measure LPO in a single cell, DLF was found to increase LPO production in H7402 cells. Conclusion: DLF can kill H7402 cells, and part of its mechanism may be through induction of tumor cells to produce NO and LPO.
