目的比较分析rIL-2刺激对人脐带血和成人外周血淋巴细胞亚群及功能的影响。方法利用流式细胞技术对脐带血和成人外周血淋巴细胞亚群进行检测,用t检验做统计学分析。结果新鲜未刺激的脐带血淋巴细胞在发育和功能方面均比成人外周血幼稚;两者对rIL-2刺激的反应明显不同,脐带血的变化主要表现为CD4+和CD8+细胞亚群的成熟和记忆功能增强,即由CD45RA+向CD45RO+细胞转化,而成人外周血淋巴细胞主要为CD33-CD16+的NK细胞比例增加和CD4+和CD8+细胞亚群的CD25+细胞比例增加,而CD45RA+和CD45RO+细胞亚群无明显变化。结论人脐带血和成人外周血淋巴细胞无论在构成还是对rIL-2刺激的反应均不同
Objective To compare the effects of rIL-2 stimulation on subsets and functions of human umbilical cord blood and adult peripheral blood lymphocytes. Methods Flow cytometry was used to detect umbilical cord blood and adult peripheral blood lymphocyte subsets, using t test to do statistical analysis. Results Fresh and unstimulated umbilical cord blood lymphocytes were immature than adult peripheral blood in both development and function. The response to rIL-2 stimulation was significantly different between the two groups. The changes of cord blood mainly manifested as maturation and memory of CD4 + and CD8 + subsets The enhanced function of CD45RA + cells transformed CD45RO + cells. However, the proportion of CD33 + CD16 + NK cells in adult peripheral blood lymphocytes was increased and the proportion of CD25 + cells in CD4 + and CD8 + subsets was increased. However, there was no significant change in CD45RA + and CD45RO + . Conclusions Human umbilical cord blood and adult peripheral blood lymphocytes respond differently both to composition and to rIL-2 stimulation
