给成年雄性大鼠注射黄体生成素释放激素(LHRH)或其高活性类似物(LHRH-A),显示明显的抗性腺作用,睾丸的LH和催乳素受体丧失,雄激素合成减少伴有睾丸及附属性器官萎缩,并抑制精子生成.LHRH的这种抗生育作用至少部分是由于内源性LH大量释放,继之性腺失敏感的结果.也有一些实验提示是由于LHRH对性腺具有直接抑制作用.为证明在体对性腺有直接作用的实验常常使用去垂体的动物.考虑到去垂体本身也会影响到性腺的机能,本文在完整动物使用LH抗血清,然后观察LHRH-A对睾丸促性腺激素受体的影响,以期在尽量减少激素紊乱的条件下鉴定LHRH-A直接性腺作用的重要性.使用体重200~225g的成年雄性大鼠,一部分静脉注射0.5ml LH抗血清(马抗牛LH的抗血清),另一部分注射等量正常马血清.两小时后,分别皮下注射不同剂量(3,10,30或100ng)的LHRH-A(〔D-丝(丁叔)~6,去甘-NH_2~(10)〕LHRH-EA),48小时后处死,做睾丸促性腺激素
