目的:研究丹参注射液联合曲安奈德对口腔黏膜下纤维性变(OSF)组织中肌成纤维细胞(MFs)生物学行为的影响,为两种药物联合使用治疗OSF探明机理。方法:采用组织块培养法分离培养OSF患者病变组织中的MFs,同法分离培养健康志愿者正常颊黏膜中的成纤维细胞。以不同浓度的丹参注射液联合曲安奈德作用于MFs,采用MTT法检测细胞增殖水平,羟脯氨酸试剂盒检测细胞分泌可溶性胶原含量的变化,免疫细胞化学法检测细胞中α-SMA的表达情况。以槟榔碱提取液刺激正常成纤维细胞,保护组加用丹参注射液联合曲安奈德,同法检测其α-SMA的表达情况。结果:不同浓度的丹参注射液联合曲安奈德可明显抑制MFs的增殖水平及胶原分泌水平,并可使其α-SMA的表达水平明显较低,还可抑制槟榔提取物促进正常成纤维细胞表达α-SMA的作用。结论:丹参注射液联合曲安奈德可抑制OSF组织中MFs的部分生物学行为,可促使其向正常成纤维细胞逆分化,并能阻止正常成纤维细胞转分化为MFs,对口腔黏膜下纤维性变患者具有一定的治疗和保护作用。
Objective: To study the effect of Salvia miltiorrhiza injection combined with triamcinolone acetonide on the biological behavior of myofibroblasts (MFs) in oral submucous fibrosis (OSF) tissues, and explore the mechanism of OSF identification for the combination of two drugs. Methods: MFs in diseased tissues of OSF patients were isolated and cultured by tissue culture method. Fibroblasts in normal buccal mucosa of healthy volunteers were isolated and cultured in the same method. The different concentrations of Salvia miltiorrhiza injection combined with triamcinolone acetonide on the MFs, MTT assay of cell proliferation, hydroxyproline kit detection of cell secretion of soluble collagen content, immunocytochemical detection of cell expression of α-SMA Happening. The arecoline extract stimulated normal fibroblasts, the protective group plus Danshen injection combined with triamcinolone acetonide, the same method to detect the expression of α-SMA. Results: Salvia miltiorrhiza injection combined with triamcinolone acetonide could significantly inhibit the proliferation and collagen secretion of MFs, and significantly lower the expression of α-SMA, and also inhibited the expression of isca nut extract in normal fibroblasts The role of α-SMA. Conclusion: Salvia miltiorrhiza injection combined with triamcinolone acetonide can inhibit part of the biological behavior of MFs in OSF tissues, which can reverse its differentiation into normal fibroblasts and prevent the normal fibroblasts from transdifferentiating into MFs. Change patients have some treatment and protection.
