高尔基体磷蛋白3(Golgi phosphoprotein 3,GOLPH3)在正常细胞中是一种磷酸化的高尔基体膜蛋白,参与执行高尔基体的蛋白质分选功能。GOLPH3已报道参与多种人类癌症,研究表明GOLPH3是一个全新的致癌基因,能促进肿瘤细胞的增殖和迁移,同时也可能是肿瘤治疗的新靶点。然而,其在胰腺癌中的临床意义和生物学作用尚不清楚。该研究通过重组慢病毒表达RNAi的方法,构建稳定细胞系使GOLPH3基因在胰腺癌细胞株colo357中表达沉默,同时构建过表达GOLPH3基因的colo357稳定细胞系,通过MTT实验,伤痕愈合实验以及Transwell实验研究GOLPH3基因在胰腺癌细胞中的增殖、侵袭以及转移能力中的作用。结果显示沉默GOLPH3基因的表达可抑制人胰腺癌细胞的增殖力、迁移力和侵袭力,提示GOLPH3有可能成为潜在的肿瘤标志物和独立的预后因子。
Golgi phosphoprotein 3 (GOLPH3), a phosphorylated Golgi membrane protein in normal cells, is involved in performing the Golgi’s protein sorting function. GOLPH3 has been reported to be involved in a variety of human cancers. Studies have shown that GOLPH3 is a novel oncogene that can promote the proliferation and migration of tumor cells and may also be a new target for cancer therapy. However, its clinical significance and biological role in pancreatic cancer are not yet clear. In this study, we constructed a stable cell line by recombinant lentivirus expressing RNAi, and silenced the expression of GOLPH3 gene in the colo357 pancreatic cancer cell line. At the same time, we constructed colo357 stable cell line overexpressing GOLPH3 gene by MTT assay, wound healing assay and Transwell assay To investigate the role of GOLPH3 in the proliferation, invasion and metastasis of pancreatic cancer cells. The results showed that silence of GOLPH3 gene expression can inhibit human pancreatic cancer cell proliferation, migration and invasiveness, suggesting that GOLPH3 may become potential tumor markers and independent prognostic factors.
