目的研究cmyc反义寡核苷酸对HL60细胞中cmyc基因的表达及HL60细胞分化的影响。方法采用针对原癌基因cmyc的mRNA5’非翻译区的一个18bp的反义寡核苷酸5’d(CTTCTCGAGGCAGGAGGG)3’与人早幼粒白血病细胞系HL60细胞共培养5d,检测cmycmRNA量的变化及其对HL60细胞分化的影响。结果用NorthernBlot检测出HL60细胞cmycmRNA的含量减少;细胞形态学变化以及非特异性酯酶(NSE)染色、硝基四氮唑蓝(NBT)还原试验均提示HL60细胞出现了分化,部分细胞向粒细胞方向分化,部分细胞向单核细胞方向分化。结论cmyc反义寡核苷酸能够抑制cmyc基因的表达并能诱导HL60细胞分化。
Aim To study the effect of cmyc antisense oligonucleotide on cmyc gene expression and HL60 cell differentiation in HL60 cells. METHODS: A 18 bp antisense oligonucleotide 5 ’(CTTCTCGAGGCAGGAGGG) 3’ against the oncogene c-myc mRNA 5 ’untranslated region was co-cultured with human promyelocytic leukemia HL60 cells for 5 days to detect c The changes of mycmRNA and their effects on the differentiation of HL60 cells. Results The content of c-mycmRNA in HL60 cells was decreased by Northern blot. The cell morphological changes and non-specific esterase (NSE) staining and NBT reduction showed that HL60 cells were differentiated and some cells Granulocytes in the direction of differentiation, some cells to monocyte differentiation. Conclusion c myc antisense oligonucleotide can inhibit the expression of c myc gene and can induce HL60 cell differentiation.
