目的:研究雄激素受体(androgenreceptor,AR)与雌激素受体(estrogenreceptor,ER)在大鼠屏状核神经元中的表达及生物学意义。方法:取80只性成熟期Wistar大鼠,雌雄各半。随机分为A、B、C、D、E、F、G、H、I、J组。用免疫组织化学染色方法检测AR、ER在屏状核神经元中的表达,四格表确切概率法行χ2检验分析。结果:AR在大鼠屏状核神经元中的阳性表达率,在D组(去势雄性雄激素干预)与F组(去势雄性)、H组(正常雄性雄激素干预)与J组(正常雄性)、F组与J组、G组(正常雌性雄激素干预)与I组(正常雌性)、F组与E组(去势雌性)之间差异有统计学意义(P<0.05);余各组间差异无统计学意义(P>0.05)。ER在大鼠屏状核神经元中的阳性表达率,在E组与I组、E组与A组(去势雌性雌激素干预)、G组与I组之间差异有统计学意义(P<0.05);余各组间差异无统计学意义(P>0.05)。结论:(1)鼠脑屏状核神经元中AR、ER均有表达;(2)大鼠屏状核为雌、雄激素的靶组织之一,雌、雄激素可能通过其受体作用于屏状核神经元而发挥其生物学效应;(3)雌、雄激素对ER、AR表达的影响在雌、雄两性可能有不同之处;(4)屏状核可能在对性冲动的唤醒及性行为中起着重要的内分泌作用。
Objective: To study the expression and biological significance of androgen receptor (AR) and estrogen receptor (ER) in the NPN neurons of rats. Methods: Eighty Wistar rats of sexual maturity were randomly divided into male and female. Randomly divided into A, B, C, D, E, F, G, H, I, J group. Immunohistochemical staining was used to detect the expression of AR and ER in NPs. The exactness of four tables was determined byχ2 test. Results: The positive expression rate of AR in rat NPs was significantly higher in group D (castration male androgen) than in group F (castration male), group H (normal male and male) and group J Normal male), F group and J group, G group (normal female androgen intervention) and I group (normal female), F group and E group (ovariectomized female), the difference was statistically significant (P <0.05); There was no significant difference among the groups (P> 0.05). The positive expression rate of ER in rat NPs was significantly different between E group and I group, E group and A group (castration female estrogen intervention), G group and I group (P <0.05). There was no significant difference among the groups (P> 0.05). Conclusions: (1) AR and ER are both expressed in the neurons of the brainstem nuclei in rats; (2) The nucleus accumbens is one of the target tissues of estrogen and androgen, and estrogen and androgens may act on (3) The effects of estrogen and androgens on the expression of ER and AR may be different in the female and male genitalia; (4) The nucleus of the striatum may wake up in sexual impulses And sexual behavior plays an important endocrine role.
