目的:建立一种简便、可靠、快速的肿瘤药敏试验新方法。方法:采用组织块培养、MTT染色和计算机图像分析法测定药物对肿瘤细胞的作用;测定加MTT后不同时间显色反应及非特异性染色以确定试验条件;对同一份标本进行3次试验以观察重复性;对人癌裸鼠移植瘤进行药敏试验以评价本试验鉴别敏感/耐药的能力;对127份临床肿瘤标本进行药敏试验,观察试验成功率。结果:加MTT反应时间以3h为合适,试验变异系数为9.4%,本试验可鉴别移植瘤的耐药性,对临床肿瘤标本试验成功率90.6%。结论:组织块培养、MTT染色和计算机图像分析的肿瘤药敏系统具有简便、可靠、快速的特点,可指导临床个体化肿瘤化疗。
Objective: To establish a simple, reliable and rapid new method of tumor susceptibility testing. Methods: The effects of drugs on tumor cells were determined by tissue culture, MTT staining and computer image analysis. The chromogenic reaction and nonspecific staining were determined at different time after addition of MTT to determine the experimental conditions. The same specimen was observed 3 times Repeatability; Human cancer xenograft tumor drug sensitivity test to evaluate the test to identify sensitive / resistant ability; 127 clinical tumor specimens susceptibility testing, the test success rate. Results: The MTT reaction time was 3h, the coefficient of variation was 9.4%. The drug resistance of xenograft tumor was identified in this experiment. The success rate of clinical tumor specimens was 90.6%. Conclusion: Tumor drug sensitivity system of tissue culture, MTT staining and computer image analysis is simple, reliable and rapid, which can guide clinical individualized tumor chemotherapy.
