斯伐他丁(simvastatin,SIM)是一种新型的羟基-β-甲基-戊二酸单酰辅酶 A 还原酶抑制剂,具有显著的降低血中胆固醇和甘油三酯的作用,但目前该药对甘油三酯的作用机理尚不清楚。选择患有高胆固醇血症及非胰岛素依赖性糖尿病的老年受试者12名,实验前3周内受试者不得服用降血糖药或其它药物,每日饮食含5.2mmol 胆固醇及至少250g 碳水化合物。采用随机、双盲交叉实验。两组分别给SIM 30mg/d 及安慰剂,持续3周,停药3周后,互换药物。每次给药3周后禁食过夜(至少12h),再随机进行:(1)口服75g 葡萄糖耐量试验(OGTT);(2)血糖正常的高胰岛素血症的葡萄糖维持实验(进行120 min)。实验表明,较之安慰剂,SIM 能显著降低血浆总胆固醇(7.9对5.3 mmol·L~(-1),p<0.001)、LDL 胆固醇(7.2对4.3mmol·L~(1-),p<0.001)、甘油三酯(2.9对2.1
