[目的 ]建立一种简便、有效的 HIV感染检测方法 ,用于儿童 HIV垂直感染的早期诊断及可疑感染者的确认。 [方法 ]用肝素化全血 (每孔分别 10 0 0、 5 0 0、 2 5 0、 12 5、 6 2 .5、 10 μl)与正常人激活的 PBMC10 6在 2 4孔板中进行复孔共培养或与新鲜的正常人 PBMC同时刺激共培养来分离 HIV病毒 ,培养上清液用 P2 4抗原 EL ISA试剂盒做检测。 [结果 ]4例全血培养中分离出 3株 HIV- 1,微量全血培养出现阳性的时间顺序为 10 μl>6 2 .5 μl>12 5 μl>2 5 0 μl>5 0 0 μl>10 0 0 μl。 [结论 ]微量全血培养有较高的敏感性和特异性 ,能够用少量全血做出快速诊断 ,可应用于 HIV- 1垂直感染的早期诊断。
[Objective] To establish a simple and effective HIV infection detection method for the early diagnosis of HIV vertical infection in children and the confirmation of suspected infection. [Method] The normal human activated PBMC10 6 was reconstituted in 24-well plates with heparinized whole blood (100, 500, 250, 125, 62.5, 10 μl per well) HIV co-culture or co-culture with fresh normal human PBMC to isolate the HIV virus, and the culture supernatant was tested with the P2 4 antigen EL ISA kit. [Results] Three HIV-1 isolates were isolated from 4 whole blood cultures, and the sequence of positive results in whole blood was 10 μl> 6 2 .5 μl> 125 μl> 250 μl> 500 μl> 10 0 0 μl. [Conclusion] The micro-whole blood culture has a high sensitivity and specificity, which can make rapid diagnosis with a small amount of whole blood and can be used in the early diagnosis of vertical HIV-1 infection.
