目的:用HPLC法建立重庆产杜仲叶的指纹图谱分析方法。方法:采用色谱柱PLATISIL ODS色谱柱(250 mm×4.6 mm,5.0μm);甲醇-0.2%H3PO4为流动相梯度洗脱,体积流量为1.0 mL/min;检测波长260 nm;进样体积20μL;柱温30℃。结果:共有13个特征峰,采用《中药色谱指纹图谱相似度评价系统(2004A版)》,计算各被测样品的HPLC指纹图谱的整体相似度,10批被测样品的色谱指纹图谱的整体相似度在0.883~0.990,并得出了重庆产杜仲叶的对照指纹图谱。结论:该指纹图谱方法简便、重现性好,为有效地控制重庆产杜仲叶的内在质量提供了科学依据。
OBJECTIVE: To establish a fingerprint method for producing Eucommia leaf in Chongqing by HPLC method. Methods: The mobile phase was eluted with a column of PLATISIL ODS (250 mm × 4.6 mm, 5.0 μm). The mobile phase consisted of methanol and 0.2% H3PO4 with a volume flow of 1.0 mL / min. The detection wavelength was 260 nm and the injection volume was 20 μL. Column temperature 30 ℃. Results: A total of 13 characteristic peaks were obtained. The overall similarity of the HPLC fingerprints of each tested sample was calculated using the “similarity evaluation system of chromatographic fingerprints of traditional Chinese medicines (2004A version)”. The overall similarity of the chromatographic fingerprints of 10 tested samples Degree of 0.883 ~ 0.990, and obtained the Chongqing produced Eucommia leaf control fingerprints. Conclusion: The fingerprinting method is simple, reproducible, and provides a scientific basis for effectively controlling the inherent quality of Eucommia leaf in Chongqing.
