目的检测胰腺癌组织和正常胰腺组织中DPC4/Smad4基因的表达差异,探讨其在胰腺癌发病机制中的作用。方法分别运用RT-PCR和免疫组化技术检测40例胰腺癌和36正常胰腺组织中的DPC4mRNA及蛋白的表达。结果胰腺癌组织中DPC4阳性16例(40%);正常胰腺组织的DPC4的阳性表达率100%(36/36)。免疫组化显示癌组织中的弥漫阳性数为4例,局灶阳性数为10例,阳性表达率35%(14/40);正常胰腺组织的弥漫阳性数为16例,局灶阳性数为20例,阳性表达率100%(36/36)。两者比较有显著性差异(P<0.05)。结论DPC4基因在胰腺癌组织中有明显的表达异常,可能与胰腺癌的发病有一定的关系,可考虑作为胰腺癌患者一个临床诊断或预后的指标。
Objective To detect the differential expression of DPC4 / Smad4 gene in pancreatic cancer tissues and normal pancreatic tissues and to explore its role in the pathogenesis of pancreatic cancer. Methods RT-PCR and immunohistochemistry were used to detect the expression of DPC4 mRNA and protein in 40 cases of pancreatic cancer and 36 normal pancreatic tissues respectively. Results The positive rate of DPC4 in pancreatic carcinoma was 16% (40%). The positive rate of DPC4 in normal pancreas was 100% (36/36). Immunohistochemistry showed that the number of diffuse positive in cancer tissue was 4, the number of focal positive was 10, and the positive expression rate was 35% (14/40). The number of diffuse positive in normal pancreas was 16, and the positive number was 20 cases, the positive expression rate of 100% (36/36). There was a significant difference between the two (P <0.05). Conclusion DPC4 gene is obviously expressed in pancreatic cancer tissues and may be related to the pathogenesis of pancreatic cancer. It may be considered as a clinical diagnostic or prognostic indicator of pancreatic cancer.
