目的观察并分析不同染毒剂量百草枯(PQ)中毒大鼠肾组织病理表现,寻找观察百草枯中毒大鼠肾损伤的最佳染毒剂量。方法SD大鼠腹腔注射不同剂量PQ,观察不同时间点肾组织病理表现,并计算实验大鼠的死亡率。结果各染毒组病理损伤在染毒后第1d即出现,以皮质部近曲小管受累最常,同一染毒组在不同时间点病理损伤的严重程度无明显差别。15mg/kg和20mg/kg染毒组可见皮质部小管上皮细胞肿胀,部分空泡变性;35mg/kg染毒组皮质部小管上皮细胞细胞核固缩增多,上皮细胞结构消失,少数肾小球结构紊乱,髓质部小管细胞核固缩,管腔消失。25mg/kg和30mg/kg染毒组病理损伤程度介于上述两组之间。随染毒剂量增加,实验大鼠病死率升高;25mg/kg染毒组死亡率为48.7%、30mg/kg染毒组死亡率为68.4%。结论25mg/kg染毒剂量可能是观察百草枯中毒大鼠肾损伤的最佳染毒剂量。
Objective To observe and analyze the pathological manifestations of kidney in rats exposed to different doses of paraquat (PQ), and to find out the best dose for observing the damage of kidney caused by paraquat poisoning in rats. Methods SD rats were injected intraperitoneally with different doses of PQ. The pathological changes of renal tissues were observed at different time points, and the mortality of experimental rats was calculated. Results The pathological changes of each exposure group appeared on the first day after exposure to the proximal tubule of the cortex. The severity of pathological injury in the same exposure group at different time points showed no significant difference. At 15mg / kg and 20mg / kg, the cortical tubule epithelial cells were swollen and partially vacuolar degeneration. In the 35mg / kg group, the cortical tubule epithelial cell nucleus pyknosis increased, the epithelial cell structure disappeared and a few glomeruli were disorganized , Medullary tubule nuclear condensation, disappearance of the lumen. The levels of pathological damage in the 25 mg / kg and 30 mg / kg groups were between the above two groups. With the increase of exposure dose, the experimental rat mortality increased; the mortality of 25mg / kg was 48.7%, and that of 30mg / kg was 68.4%. Conclusion The dose of 25mg / kg may be the best dose to observe the injury of paraquat in rats.
