为寻找新的肿瘤细胞诱导分化剂,采用MTT比色分析法和形态定量分析技术检测了淫羊藿甙对人急性早幼粒白血病细胞(HL-60)增殖与分化的影响。结果表明:淫羊藿甙可以抑制HL-60细胞的增殖,呈剂量依赖关系;选择62.5、125、250mg·L-1进行动态观察,发现12h后即可发挥抑制作用。淫羊藿甙100mg·L-1作用48h后HL-60细胞NBT还原能力明显增强(P<0.01),细胞核面积减小,平均光密度(MOD)增加,积分光密度减小(IOD);光镜观察结果显示,淫羊藿甙作用后的HL-60细胞发生明显变化,细胞核体积减小,细胞核呈杆状或分叶状。上述结果提示,淫羊藿甙对HL-60细胞有诱导分化作用。
In order to find new tumor cell differentiation inducer, the effect of epimedium on the proliferation and differentiation of human acute promyelocytic leukemia cells (HL-60) was detected by MTT assay and morphometric quantitative analysis. The results showed that epimedium can inhibit the proliferation of HL-60 cells in a dose-dependent manner; 62.5, 125, and 250 mg·L-1 were selected for dynamic observation, and it was found that inhibition was achieved after 12 hours. After treatment with Epimedium 100 mg·L-1 for 48 h, the reducing ability of NBT in HL-60 cells was significantly increased (P<0.01), the nucleus area was decreased, the average optical density (MOD) increased, and the integrated optical density decreased (IOD). The results of light microscopy showed that the epidermal growth of HL-60 cells significantly changed, the nuclei volume decreased, and the nuclei were rod-shaped or lobulated. The above results suggest that epimedium induces differentiation of HL-60 cells.
