目的 了解慢性乙型肝炎病毒(HBV) 感染者外周血单个核细胞(PBMC) 中CD4 + T 细胞内T辅助细胞1、2 型(Th1/Th2) 因子、干扰素γ(IFNγ) 和白细胞介素4(IL4) 的表达情况,以测定Th0/Th1/Th2 细胞的百分数,探明Th1/Th2 细胞在慢性HBV 感染中的作用。方法 常规分离PBMC,在佛波二酯(PMA)、Ionomycin 、Monensin 的刺激下,采用流式细胞检测仪(FACS) 对慢性HBV感染者外周血单个CD4+ T 细胞内IL4 和IFNγ表达进行分析。结果 正常对照组,2 .3 % ~18 .6 %的CD4 + T 细胞为Th1 细胞,0 .9 % ~9 .2% 为Th2 细胞;在慢性HBV 感染者外周血单个CD4 + T 细胞中,以Th0 细胞居多;而Th1 细胞则随着慢性乙型肝炎肝脏炎症活动的加剧而明显增多,在炎症活动明显的慢性乙型肝炎,其Th1 百分数明显高于炎症活动呈静止状态的慢性乙型肝炎;Th2 细胞在慢性HBV 感染的不同阶段则较为恒定,但明显高于对照组。结论 Th1 细胞与炎症活动呈正相关,Th2 细胞可能与HBV 感染的慢性化相关联
Objective To investigate the expressions of Th1 / Th2 and IFNγ in CD4 + T cells in peripheral blood mononuclear cells (PBMCs) from patients with chronic hepatitis B virus infection Interleukin 4 (IL 4) expression, to determine the Th0 / Th1 / Th2 cell percentage, to explore Th1 / Th2 cells in chronic HBV infection in the role. Methods PBMCs were isolated routinely and stimulated by PMA, Ionomycin and Monensin to detect the expression of IL4 and IFNγ in CD4 + T cells from peripheral blood of patients with chronic HBV infection by flow cytometry (FACS) Analyze. Results In the normal control group, 2.3% ~ 18.6% of CD4 + T cells were Th1 cells and 0.9% ~ 9.2% of them were Th2 cells. In peripheral blood CD4 + T cells of chronic HBV infected persons, While Th1 cells increased significantly with the increase of hepatic inflammatory activity of chronic hepatitis B. The Th1 percentage of chronic hepatitis B with obvious inflammatory activity was significantly higher than that of chronic hepatitis B with inflammatory activity ; Th2 cells in the different stages of chronic HBV infection is more constant, but significantly higher than the control group. Conclusion There is a positive correlation between Th1 cells and inflammatory activity and Th2 cells may be associated with chronic HBV infection
