以水提醇沉法获得“沪农灵芝1号”子实体中的灵芝多糖,并对其结构特征和含量进行分析。高效凝胶尺寸排阻色谱-多角度激光光散射仪-示差折光检测仪联用分析结果表明,该多糖重均相对分子质量为2.28×106,均方根半径为150.1 nm,多分散系数(Mw/Mn)为1.14,在水溶液中呈棒状结构。单糖组成及甲基化分析结合酶解试验结果表明,该多糖为β-1,3/1,6-葡聚糖。经液相色谱法定量检测,段木栽培所得灵芝子实体中,该多糖质量分数为0.52 mg/g,显著高于袋料栽培所得子实体中相应组分的含量。
Water-alcohol precipitation method to obtain “Shanghai Farrell Ganoderma lucidum No. 1” fruiting bodies of Ganoderma lucidum polysaccharides, and its structural characteristics and content were analyzed. The results of high performance gel exclusion chromatography-multi-angle laser light scattering and refractive index detector showed that the weight average molecular weight of this polysaccharide was 2.28 × 106 and the root mean square radius was 150.1 nm. The polydispersity (Mw / Mn) of 1.14, showing a rod-like structure in an aqueous solution. Monosaccharide composition and methylation analysis combined with enzymatic hydrolysis test results show that the polysaccharide is β-1,3 / 1,6-glucan. Quantitative detection by liquid chromatography, segment wood cultivation Ganoderma lucidum fruiting body, the polysaccharide mass fraction of 0.52 mg / g, significantly higher than the bag-grown fruiting body corresponding component content.
