目的:运用Real time PCR技术观察益气破血解毒小复方干预对NZB/WF1狼疮鼠脾淋巴细胞IFN-γ、IL-10基因表达的影响。方法:58只5月龄雌性NZB/WF1狼疮鼠随机分配至3组:空白组20只、中药组20只和西药组18只。空白组灌胃无菌生理盐水0.5ml/d,中药组灌胃益气破血解毒小复方0.5ml/d,西药组灌胃骁悉溶液0.5ml/d,中药组灌胃益气破血解毒小复方0.5ml/d,西药组灌胃骁悉溶液0.5ml/d(浓度1.3mg/ml),干预时间为60天。无菌条件下取出小鼠脾脏,机械分离、梯度离心法分离淋巴细胞。恒温培养72h后收获细胞,抽提总RNA,用Real time PCR法行IFN-γ、IL-10基因表达检测。结果:空白组的IFN-γ基因表达量显著高于西药组(P<0.01),而与中药组间无统计学差异(P>0.05)。空白组IL-10基因表达量显著高于中药组(P<0.05)和西药组(P<0.05),两治疗组间未见统计学差异(P>0.05)。
OBJECTIVE: To observe the effect of Yiqi Poxue Xiaodu Compound Prescription on gene expression of IFN-γ and IL-10 in splenic lymphocytes of NZB/WF1 lupus erythematosus mice using Real time PCR. Methods: A total of 58 5-month-old female NZB/WF1 lupus mice were randomly assigned to 3 groups: 20 in the blank group, 20 in the Chinese medicine group and 18 in the western medicine group. The blank group was given intragastric sterile saline 0.5ml/d. The Chinese medicine group was given Yiqi Decoction, small compound 0.5ml/d, the Western medicine group was intragastrically administered 0.5ml/d, and the Chinese medicine group was given Yiqixuexuejiedu. The small compound 0.5ml/d, Western medicine group gavage solution 0.5ml/d (concentration 1.3mg/ml), the intervention time is 60 days. The spleen of the mice was aseptically removed, and lymphocytes were isolated by mechanical separation and gradient centrifugation. The cells were harvested after constant temperature culture for 72 hours, total RNA was extracted, and IFN-γ and IL-10 gene expression was detected by Real time PCR. Results: The expression level of IFN-γ gene in the blank group was significantly higher than that in the western medicine group (P<0.01), but there was no statistical difference between the blank group and the traditional Chinese medicine group (P>0.05). The expression level of IL-10 gene in blank group was significantly higher than that in Chinese medicine group (P<0.05) and Western medicine group (P<0.05). There was no statistical difference between the two treatment groups (P>0.05).
