目的 观察锌对缺碘大鼠甲状腺细胞膜脂流动性的影响。方法 应用缺碘饲料喂养 Wistar大鼠复制缺碘动物模型 ,同时设补碘、补葡萄糖酸锌和复合锌组。实验中测尿碘 ,实验结束时测血清、组织锌含量 ;以DPH为荧光探剂 ,荧光偏振法测甲状腺细胞膜脂荧光偏振度 (P)和膜相对流动性 (MFU)。结果 缺碘组尿碘降低 ,加碘组和两组加锌组尿碘、组织锌升高。缺碘组甲状腺细胞膜脂 P值升高 ,MF U降低 ;加碘组 P值和 MFU基本达对照组水平 ;两组加锌组较缺碘组 P值降低 ,MF U升高。结论 缺碘大鼠甲状腺细胞膜脂流动性降低 ,补锌可拮抗缺碘所致甲状腺细胞膜损伤 ,但作用不及补碘显著
Objective To observe the effect of zinc on the membrane fluidity of thyroid cells in iodine deficiency rats. Methods Iodine-deficient diet was used to feed iodine deficient animal model in Wistar rats. At the same time, iodine, zinc gluconate and zinc complex were set up. The urinary iodine was measured in the experiment. The serum and tissue zinc contents were measured at the end of the experiment. The fluorescence polarization (P) and membrane relative fluidity (MFU) of the membrane lipids of the thyrocytes were measured by fluorescence polarization method with DPH as fluorescent probe. Results The iodine deficiency group decreased urinary iodine, iodine group and two groups of zinc plus urine iodine, zinc tissue. In the iodine-deficient group, the P value of thyroid cells increased and the MFU decreased. The values of P and MFU in the iodized group reached the level of the control group. The P value and the MFU of the two groups were lower than those of the iodine-deficient group. Conclusions The iodine-deficient rat thyroid cell membrane lipid fluidity is reduced, and zinc supplementation can antagonize the thyroid cell membrane damage caused by iodine deficiency, but the effect is not as significant as iodine supplement
