目的 :探讨同型半胱氨酸 (Hcy)对血管平滑肌细胞 (VSMC)生长的影响。方法 :采用MTT法检测细胞数量及流式细胞术测定细胞周期。结果 :5× 10 -4mol/LHcy干预处于生长静止期的细胞 4 8、72h细胞数量较对照组明显增加 (P <0 .0 1) ,分别增加了 5 0 .75 %和 84 .83%。当采用 1× 10 -4,2 .5× 10 -4,5× 10 -4,1× 10 -3mol/L的Hcy干预细胞 72h后 ,其细胞数量分别较对照组增加了4 1.5 6 % ,5 8.4 4 % ,6 6 .2 3% ,74 .0 2 %。并且当 5× 10 -4mol/LHcy干预细胞 2 4、36、4 8h后 ,其S期、G2 /M期在细胞周期的比例也明显增加 (P <0 .0 1)。结论 :Hcy能明显诱导VSMC增殖 ,并呈现时间和剂量依赖关系 ,提示Hcy通过诱导VSMC的增殖可导致动脉粥样硬化的发生。
Objective: To investigate the effect of homocysteine (Hcy) on the growth of vascular smooth muscle cells (VSMCs). Methods: The cell number was determined by MTT assay and the cell cycle was determined by flow cytometry. Results: The number of cells in quiescent cells at 48 h and 72 h after 5 × 10 -4 mol / LHcy intervention was significantly increased compared with the control group (P <0.01), increasing by 50.7% and 84.83% respectively. When Hcy interventional cells at 1 × 10 -4,2. 5 × 10 -4, 5 × 10 -4 and 1 × 10 -3 mol / L were used for 72 h, their cell numbers increased by 41.56% 5 8.4 4%, 6 6 .2 3%, 74 .0 2%. The ratio of S phase and G2 / M phase in the cell cycle was also significantly increased after 5 × 10 -4 mol / LHcy intervention for 24, 36 and 48 hours (P <0.01). CONCLUSION: Hcy can obviously induce the proliferation of VSMCs in a time-and dose-dependent manner, suggesting that Hcy can induce the development of atherosclerosis by inducing VSMC proliferation.
