[Objective]To establish a chronic alcoholic liver injury model by observing mice and explore the main mechanism of the vaccinium oxycoccus pigment protecting the liver from alcoholic injuries in the perspective of anti-oxidation.[Methods]Randomly divide the experimental into four groups (A: normal control group B: Vaccinium Oxycoccus Pigment group;C: only ethanol (EtOH) group;D: EtOH + Vaccinium Oxycoccus Pigment group);expose C and D groups to gradient amounts of alcohol for consecutive six weeks (f 2.4 g / kg / d in first four weeks ago and then increase it to 3 g / kg / d in subsequent two weeks) to establish a chronic alcoholic liver injury model;subject B and D groups to the intervention of the vaccinium oxycoccus pigment of 50 mg / kg / d;after the experiment,collect the blood and liver tissues,make pathological slices of the liver tissues and measure the levels of serum transaminases (AST,ALT),lipid metabolism (TC,TG,HDL-C),ROS in hepatic tissues and oxidative / anti-oxidative systems (T-AOC,GSH,GPx,GST,MDA).[Results]Oxidative injuries of alcohol to liver cells were positively correlated with the concentration of alcohol and the exposure time.The vaccinium oxycoccus pigment could reduce alcohol-induced oxidative liver injuries by mitigating hepatic steatosis,inhibiting reduced transaminase and slowed cellular metabolism,regulating lipid metabolism disorder,suppressing ROS accumulation in the liver,inducing HO-1 activity and expression in liver cells and prompting the early restoration of imbalanced anti-oxidative system.[Conclusion]The vaccinium oxycoccus pigment can reduce alcohol-induced oxidative liver injuries.
