1,5-Benzodiazepines,very important compounds,are widely used in the last decades as anticonvulsant,antianxiety and hypnotic agents.[1]In recent years,1,5-benzodiazepine and its derivatives have been also reported to exhibit excellent antimicrobial activity.Moreover,some researches have reported that some compounds with lactone showed high biological activity,such as anti-feeding,anti-diabetic,especially anti-bacterial and anti-fungal.[2]Therefore,we designed and synthesized a series of unreported novel substituted 2-phenyl-1,5-benzodiazepine-4-propiolactone compounds in two steps (scheme 1).The intermediate products Ⅰ were obtained via the condensation reaction of ethyl acetoacetate with 3-substituted benzaldehyde in EtOH in the presence of NaOH for 1.5h under reflux.Then intermediate products Ⅰ and substituted o-phenylenediamine was stirred without any catalyst in EtOH at ice-bath to obtain the target compounds Ⅱ with excellent yields (85%-93%).Simple operation,short reaction time,high yields are the advantages of this method.The target compounds were determined by 1H NMR,13C NMR,MS,IR and elemental analysis.Such compounds lay a good foundation for the study of 1,5-benzodiazepine lactone compounds in the mechanism,the synthetic route and the method.
