目的:研究娑罗子总皂苷的主要活性成分escin Ⅰa对三阴性乳腺癌(triple-negative breast cancer,TNBC)转移的抑制作用,并探讨可能的机制.方法:采用transwell法测定MDA-MB-231的侵袭;Q-PCR法测定BRMS1、E-cadherin、Keratin19、LOXL2、MMP9、Orai1、Stim1、VEGF、TGFβ等侵袭相关因子和EMT关键转录因子Snail、Slug、Zeb1、Zeb2、Twist的mRNA表达;Western Blot法测定HIF-1 α、LOXL2及EMT标志蛋白E-cadherin、vimentin和α-SMA的表达.此外,建立MDA-MB-231异种移植瘤模型,考察escin Ⅰa对MDA-MB-231异种移植瘤肺转移及原位肿瘤HIF-1 α、LOXL2及EMT标志蛋白的影响.
