A simple and fast HPLC analysis method for cocaine HCl in cocaine sample was developed. In this work, cocaine in sample was separated using a Shim-pack VP-ODS column (250 mm × 4.6 mm, 5 μm) and a mobile phase of methanol-0.05% TFA (35:65, v/v) at 35 ℃. The flow rate of a mobile phase was set to 1.0 mL/min and the UV detection was performed at 231 nm. Our results show that there was good linearity in the range of 1-1000 μg/mL of cocaine HCl concentration (R2=0.9998, n=6). The average recovery rate was 99.71% (n=9, RSD=0.54%). The limit of detection was found to be ~0.05 μg/mL.
