目的观察七龙天对实验性肺动脉高压(PAH)大鼠内皮素1(ET-1)、白三烯C4(LTC4)的影响。方法雄性SD大鼠随机分为6组:空白对照组(A)、模型组(B)、硝苯吡啶组(C)、七龙天低剂量组(D)、七龙天中剂量组(E)、七龙天高剂量组(F)。空白对照组大鼠以等量0.9%NaCl溶液腹腔注射,其余各组均以野百合碱(MCT)腹腔注射复制PAH模型造模。制模后14天开始,各组分别定时灌胃给药,每日1次,连续给药8天至检查前12h停药。以改良右心导管术测定肺血流动力学参数,用放射免疫(RIA)法测定血浆ET-1,酶联免疫吸附法(ELISA)测定血清LTC4的变化。结果与A组比较,其余各组大鼠PASP、PADP、MPAP、ET-1、LTC4水平较空白对照组均有所升高,且B组升高最显著(P<0.01);与B组比较,C、D、E、F组均可降低大鼠PASP、PADP、MPAP、ET-1水平(P<0.05,P<0.01);与C、D组比较,E、F组大鼠PASP、PADP、MPAP、ET-1水平降低更明显(P<0.05);各用药组对LTC4的影响差异无统计学意义(P>0.05)。结论七龙天能够有效降低实验动物的肺动脉压,其机制与降低ET-1含量有关。
Objective To observe the effect of “Chinese Dragon” on endothelin 1 (ET-1) and leukotriene C4 (LTC4) in rats with experimental pulmonary hypertension (PAH). Methods Male Sprague-Dawley rats were randomly divided into 6 groups: control group (A), model group (B), nifedipine group (C), Danshitian low dose group ), High-dose Tianlongtian dose group (F). The rats in blank control group were injected intraperitoneally with 0.9% NaCl solution in equal volume. The other groups were given intraperitoneal injection of monocrotaline (MCT) to replicate PAH model. 14 days after the model began, each group were given gavage regularly, once daily, continuous administration of 8 days to check before 12h withdrawal. Pulmonary hemodynamics parameters were measured by modified right heart catheterization, plasma ET-1 was measured by radioimmunoassay (RIA) and serum LTC4 by enzyme-linked immunosorbent assay (ELISA). Results Compared with group A, the levels of PASP, PADP, MPAP, ET-1 and LTC4 in rats in other groups were significantly higher than those in control group (P <0.01), and those in group B were the highest (P <0.01) PASP, PADP, MPAP and ET-1 in rats in groups C, D, E and F decreased significantly (P <0.05, P <0.01) (P <0.05). There was no significant difference in the effect of each treatment group on LTC4 (P> 0.05). Conclusion Seven Dragons can effectively reduce the pulmonary arterial pressure in experimental animals, the mechanism of which is related to the decrease of ET-1 content.
