Determination the Changes of Saponin Compositions by High Temperature Steaming Process with HPLC-MS

  Ginseng(Panax ginseng C.A.Meyer)is chinese traditional medicine.Ginseng saponin is the main functional component which can effectively enhance the central nervous,cardiovascular protection,and increase human immunity[1].However there is a few of reports about the changes of the ginseng saponins after high temperature processing as yet.The optimal methods for extraction of the total ginseng saponins,through observing the influences of extraction temperature and extraction time on extraction yield,were obtained by using orthogonal test in this paper.The total ginseng saponin content was analyzed by using ultraviolet-visible spectrophotometry,and the compositions changes of ginseng saponins before and after high temperature treatment were measured by using high performance liquid chromatography(HPLC)and mass chromatography[3,4].The results showed that during the period of steaming,the sugar base of ginseng saponin was taken off on C-20 firstly,then elimination was occur on C-6 or C-3,next dehydration was occur on C-20,finally the unsaturated ginseng saponin was generated.After steaming,the contents of Rb1,Rc,Rb2 and Rd in ginseng were reduced,generating a little of the ginseng saponin 20S-Rg3,20R-Rg3,Rkl and Rg5.Studies have shown that secondary ginsenoside Rg3 and 20s-Rg3 have a strong activity on anti-tumor and can enhance immunity; Rk1 has better effects on improving memory; Rg5 has antitumor effect[2].Above the substances possessing important pharmacological activities generated during high temperature process could provide the bases for the development and utilization of ginseng saponin in clinic.
