目的:研究峨眉桃叶珊瑚果实中桃叶珊瑚苷的提取纯化工艺.方法:以提取率为指标,采用单因素法考察峨眉桃叶珊瑚果实中桃叶珊瑚苷醇提工艺;以纯度和回收率为指标,考察桃叶珊瑚苷的硅胶柱色谱法及结晶法纯化工艺.结果:桃叶珊瑚苷的最佳提取条件:以80%乙醇为溶剂,料液比1:10,提取温度80℃,回流提取3次,1.0h/次;硅胶柱色谱纯化工艺:以甲醇-氯仿(1∶5和1∶3)梯度洗脱,洗脱剂用量为3 BV,柱子径高

目的:研究峨眉桃叶珊瑚果实中桃叶珊瑚苷的提取纯化工艺.方法:以提取率为指标,采用单因素法考察峨眉桃叶珊瑚果实中桃叶珊瑚苷醇提工艺;以纯度和回收率为指标,考察桃叶珊瑚苷的硅胶柱色谱法及结晶法纯化工艺.结果:桃叶珊瑚苷的最佳提取条件:以80%乙醇为溶剂,料液比1:10,提取温度80℃,回流提取3次,1.0h/次;硅胶柱色谱纯化工艺:以甲醇-氯仿(1∶5和1∶3)梯度洗脱,洗脱剂用量为3 BV,柱子径高比为1∶15,流速为2 BV·h-1,上样量35 mg·mL-1;结晶工艺:以正丁醇为结晶溶剂,加热沸腾使样品完全溶解,40℃保温过滤,滤液减压回收正丁醇至20倍量,低温搅拌析晶2h,得粉末状晶体后,低温冷冻离心3次,取下层真空干燥即得桃叶珊瑚苷精制品,其纯度与回收率的平均值约为93.07%与70.94%.结论:该工艺可较好提取纯化峨眉桃叶珊瑚果实中桃叶珊瑚苷.

本文通过Alcalase蛋白酶和Protamex蛋白酶对灵芝蛋白提取液进行水解并对水解前后的灵芝蛋白的抗氧化活性进行了研究.研究结果表明,灵芝蛋白提取液与酶解液均具有抗氧化活性,随着灵芝蛋白浓度增加,蛋白提取液清除·oH自由基的能力和还原力都增强,当灵芝蛋白的浓度为0.28mg/mL时,蛋白提取液的·OH自由基清除率为28.7%,且清除·OH自由基的能力与灵芝蛋白的浓度的关系可以用对数方程来描述.
