Glutamate uptake triggers astrocytic nuclear swelling through aquaporin-1 located on nuclear membran

  In the central nervous system,excitatory neurotransmitter glutamate is mainly released from neuron and uptaken by astrocyte.In addition to our previous findings that glutamate in astrocyte could be synthesized into glutamine and utilized in TCA cycle for energy metabolism,we observed that glutamate induced progressive nuclear swelling in astrocyte.The rate of nuclear swelling was glutamate concentration dependent.Glutamate-induced nuclear swelling was also observed in isolated astrocytic nucleus,indicating nuclear swelling was a direct effect and independent from cytosolic components.Glutamate-induced nuclear swelling could be modulated by glutamate transporters,but not glutamate receptors.It also required the presence of aquaporin-1,which we identified to be localized on the astrocytic nuclear envelope.Our findings indicated that glutamate in astrocyte could not only affect its metabolism,but also nuclear functions.
