We report an ultra performance liquid chromatography (UPLC) separation with evaporative lightscattering detection (ELSD) method for the rapid and direct analysis of six maltooligosacc6arides. UPLCseparation employing a mobile phase with pure water and acetonitrile containing 0.2% triethylamine resulted inenhanced separation and a running time of less than 6 minutes. Only a simple liquid-liquid extraction was requiredfor the sample pretreatment. This UPLC-ELSD method provided limits of quantifications (LOQs) from 15.0--25.0μg/mL for the six maltooligosaccharides ranging from 15 to 2000 pg/mL and showed excellent linearity(R2≥0.9914). This optimized method provided a satisfactory infra-day precision of 2.4 %-9.4 %,inter-dayprecision of 5.5 %-12.1%,accuracy of -7.7%-+11.4%at each level. We have successfully applied this method todetermine the six maltooligosaccharide concentrations in honey samples from apiaries and supermarkets.Concentrations of six maltooligosaccharides in 290 honey samples varied from undetectable to 14.1 mg/mL andthe lowest concentration that could be detected by the developed method was 0.132 mg/mL. This studydemonstrates the potential application of the UPLC-ELSD technique for sensitive monitotoring of the authenticity ofhoney samples in the food processing industry.
