为获知亚硝酸盐对团头鲂肝脏抗氧化系统胁迫机制,本实验采用同源克隆和荧光定量PCR的技术,结合氧化应激指标测定和组织学分析,系统的探讨亚硝酸盐胁迫下活性氧自由基调控团头鲂抗氧化酶的作用机制。结果显示,超氧化物歧化酶(Cu/Zn SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GPx1)基因cDNA全长分别为1575,570和573bp,分别编码525,190和191氨基酸。与其它物种抗氧化基因氨基酸序列进行同源性比较,团头鲂抗氧化氨基酸序列与鲤科鱼类紧密聚为一支。荧光定量PCR结果显示,团头鲂抗氧化基因在各组织中均有表达,其中肝脏中高度表达,团头鲂抗氧化基因在急性亚硝酸盐胁迫下,其表达模式均为先上升后下降;进一步研究表明,过量的活性氧自由基对团头鲂肝脏的脂肪和蛋白组织及抗氧化酶具有损害作用,肝细胞组织学证实亚硝酸盐胁迫导致肝细胞肿大、细胞核萎缩和形成脂肪滴。研究表明,活性氧自由基和抗氧化系统的不平衡是导致亚硝酸盐毒性胁迫机制的主要原因,氧化胁迫标志物的显著增加和肝脏组织损坏证实团头鲂抗氧化基因的转录水平显著降低的表达模式。
