基于二元电渗流泵的纳流梯度生成器及其在微型色谱中的应用

  对于微型色谱而言,梯度洗脱是增强其分离能力的重要手段[1],而双泵或四泵体系常用来构建梯度生成装置。在以往的实验中,我们发展了基于整体柱的高压电渗流泵[2]。本实验中,我们进一步优化了整体柱基电渗流泵的制备方法,并基于二元电渗流泵构建了用于微型液相色谱的纳流梯度生成器。该系统包含两个整体柱基电渗流泵,一个12通阀,一个纳升进样器,一个填充毛细管柱和一台紫外吸收检测器(Figure 1)。在电渗流泵的两端分别施加梯度电压(一升一降),两泵以变化的流速推动各自储液环内的流动相发生混合,从而形成梯度洗脱;其梯度流形是线性的,且易于调控。该系统成功应用于蛋白质消解产物的梯度分离,获得了可与商用液相色谱相比拟的分离效率和峰容量。
