The relative expression foldsofCsMYB and CsbHLH were significantly greater in the Citrus sinensis cv.Tarocco fruits,butCsWD40 transcripts were found in both blood orangeTaroccoand blonde navel orange21st Centuryby fruit transcriptome sequencing.CsMYB and CsbHLH are uniquely expressed in the fruit but the CsWD40 was found in leaf,petal,fruit,stigma and stamensof theTarroco.In particular,R2R3-MYB,bHLH-type and WD40-repeat proteins (encoded by CsMYB,CsbHLH and CsWD40) were transcription factors with conserved domain respectively and interactedin vitro and in vivo.In comparison with other two transcription factors,CsMYB transcription levels were positively correlated with anthocyanin contents in all blood orange cultivars.Moreover,the structural genesinvolved into anthocyanin biosynthesis had higher expression folds both in abud mutationTaroccofruit with deeper coloration and transgenic seedling of Arabidopsis thaliama by CsMYBoverexpression.Furthermore,CsMYB,CsbHLH and Glutathione S-transferases (GST)inTarrocowere activated by cold stimuli and then had similar expression profilingduring cold stress,respectively.In conclusion,CsbHLH,CsbHLH and CsWD40encoding proteinsmight be involved into anthocyanin regulation in the Citrus sinensis cv.Taroccofruits by an activating complex (MYB-bHLH-WD40) as well as other fruit trees.
