活性氧/活性氮(ROS/RNS)产物是虾类血细胞胞内重要的杀菌物质,研究不同类型血细胞的氧化途径,有助于进一步明确各类血细胞的免疫功能。取斑节对虾血细胞,添加抑制剂进行处理后,应用流式细胞术(FCM)测定胞内ROS/RNS含量的变化。结果 显示,透明细胞的ROS/RNS含量极低,各类抑制剂对其含量没有显著影响；经apocynin(NADPH氧化酶)处理30min后,大颗粒细胞和小颗粒细胞的ROS/RNS含量分别下降了37％和63％；经L-NMMA(一氧化氮合酶抑制剂)处理30min后,大颗粒细胞和小颗粒细胞的ROS/RNS含量分别下降了52％和30％；经DPI(同时抑制NADPH氧化酶和NO合酶)处理30min后,大颗粒细胞和小颗粒细胞的ROS/RNS含量分别下降了79％和84％。这些结果表明大颗粒细胞和小颗粒细胞是产生ROS/RNS进行杀菌的主要血细胞类型,两类血细胞均可同时产生ROS和RNS,其中大颗粒细胞主要依赖NO合酶产生NO的氧化途径,而小颗粒细胞主要依赖NADPH氧化酶产生ROS的氧化途径。
