囊胚注射TS细胞制作嵌合体胚胎生产克隆猪的研究

  滋养层干细胞(TS)是胎盘组织细胞的祖细胞,与胚胎着床和胎盘的形成密切相关。胚胎着床和胎盘形成是母体与胎儿建立联系的关键时期,此时期滋养层细胞增殖速度快,滋养层干细胞自我更新和分化旺盛,与多种妊娠相关疾病的发生、发展及其增殖、功能调节的失常有密切关系。本研究利用人工受精猪囊胚分离培养得到猪TS细胞,在此基础上,以TS细胞为供体细胞,以杜洛克克隆胚胎为受体,通过piezo系统将TS细胞注射到囊胚腔内,经过恢复培养后,移植到代孕母猪的子宫中,通过生猪情况评价该方法对克隆效率的影响。
