Non-toxic dwarf self-rooted seedling close planting cultivation is the main trend of apple development in the world.Compared with dwarfing interstock,the dwarf self-rooted rootstock is early bearing,neat,easy to manage,high yield and good fruit quality.M9-T337 is an excellent dwarf rootstock which is widely used in intensive cultivation of apple at present.Plant tissue culture is not restricted by external conditions,can be produced annually,and can maintain the excellent characteristics of the parent,is widely used in actual production.In this study,apple M9T337 dwarfing rootstock was used as experimental material.The techniques of establishing explants,proliferation medium,rooting medium and seedling transplantation were studied,so as to establish and optimize the technology system of tissue culture and rapid propagation of M9-T337.It also provides theoretical and technical support for the asexual breeding of dwarf rootstocks,and benefit to realize standardized production on a large scale,improve the self-sufficiency rate of dwarfed rootstocks,and better meet the current needs of fruit industry development.The main results are as follows:1.The most suitable disinfection method of M9-T337 is 0.1%HgCl2,and the suitable time is 8 min.The best time to establish the explants of M9-T337 is from late April to early May.The best proliferation medium is MS+6-BA 0.55 mg/L+IBA 0.75 mg/L+sucrose 40g/L.The effect of different auxin combinations is better than that of a single auxin.The suitable rooting medium is 1/2MS+IBA 0.5 mg/L+IAA 0.5 mg/L+sucrose 25 g/L,and the root length is 5.5 cm,rooting rate was 96.6%.2.The survival rate of M9-T337 self-rooted seedling was greatly influenced by the length of seedling training time,transplanting medium and seedling training method.The survival rate of transplanting was the highest at 7 days after training seedlings,reaching 94%.The survival rate can reach 97.3%when the vermiculite:matrix is 1:2.The method of closing the bottle and then opening the bottle for seedling cultivation has a higher survival rate than transplanting with the bottle closed for seedling cultivation only.The survival rate is the highest,reaching 86.6,after 10 days of bottle-closed train seedling and 5 days of bottle-opened train seedling.The best time for field transplanting is April to May,September to October,when the survival rate is the highest.
