目的 观察大鼠在孕期、哺乳期维生素D (Vitamin D)缺乏对子代鼠的肺组织结构发育及细胞周期蛋白D1 (CyclinD1)表达的影响.方法 成年雌性SD大鼠完全随机分配为维生素D缺乏模型组(VDD组)及正常对照组(对照组),每组共6只,缺D组予完全避光、不合维生素D饲料喂养,正常组予正常光照、正常饲料喂养,2周后,与成年雄性SD大鼠以2:1比例合笼交配,建立VitD孕鼠缺乏模型.利用竞争性酶联免疫吸附试验技术(ELISA)检测母鼠产后21天血清25-(OH)-D3的水平.分别取出生后第1、7、14、21天子代鼠肺组织,光镜下分别观察肺组织形态结构,免疫组化法分别检测肺组织中CyclinD1的表达.结果 VDD组雌鼠血清25-(OH)-D3水平显著低于对照组(P<0.05);光镜下同日龄VDD组肺组织结构较对照组肺泡间隔增宽,肺泡组织少,肺泡小而不规则,终末细支气管及小支气管形态不规则;免疫组化法检测CyclinD1 VDD组及对照组均在支气管黏膜上皮细胞表达明显,对照组CyclinD1在生后第1天表达最强,第7、14、21天逐渐减弱,而VDD组CyclinD1在生后第7天表达最强,高于对照组(P<0.05),第1天表达低于对照组(P<0.05),第14、21天逐渐减弱,均高于对照组,差异有统计学意义(P<0.05).结论 母鼠孕期、哺乳期维生素D缺乏延缓子代鼠的肺发育过程,CyclinD1参与肺组织细胞的增殖,CyclinD1表达延迟可能是母鼠VitD缺乏抑制子代鼠肺发育的可能机制之一.“,”Objective:To observe the effect of rats lack of Vitamin D during pregnancy and lactation on the lung development and CyclinD1 expression of offspring rats.Methods:The adult female Sprague-Dawley rats were randomly assigned to vitamin D deficiency group (VDD group) and normal control group (control group),a total of 6 rats in each group,VDD group avoided light completely and fed by fodder which do not contain vitamin D,control group had the normal illumination and general feed,2 weeks later,the female rats copulated with the adult male SD rats in the same cage with 2-1 ratio,and then we established a model of VitD deficiency of pregnant rats.By using competitive enzyme-linked immunosorbent assay (ELISA) technology to detect the levels of serum 25-(OH)-D3 of the female rats after postpartum 21 days.Respectively took lung tissue of the 1,7,14,21 days after offspring rat borned,then observed the lung tissue morphological structure by microscope respectively,at the same time,detected the expression of CyclinD1 in each lung tissue by immunohistochemical method.Results:The serum 25 (OH) D3 level of the female mices in VDD group was significantly lower than control group (P<0.05);the lung tissue structure observed on the same day of VDD group was wider alveolar septa,less alveolar tissue,smaller and irregular alveoli,irregular terminal bronchioles and small bronchi than control group;VDD group and control group each in the bronchial mucosa epithelial cells express CyclinD1 clearly,the control group expressed CyclinD1 strongest in 1st day after the birth of the newborn rats,and waned in the 7th,14th,21st days.But,the VDD group expressed CyclinD1 strongest in 7th day after birth,higher than the control group (P<0.05),the 1st day expression was lower than the control group (P<0.05),the 14th,21st days expression were higher than the control group (P<0.05).Conclusion:Vitamin D deficiency of the female rats in pregnancy or lactation period delay lung development of offspring rats,CyclinD1 participate in lung tissue cell proliferation,one of the possible mechanisms of the female rats inhibite offspring rats lung development is likely to be vitamin D deficiency delay expression of CyclinD1.
