目的:了解汉族人群瘦素受体Gln223Arg基因型分布;探讨该基因多态性与胃癌的关系。方法:运用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法测定无亲缘关系的221例汉族人群的瘦素受体基因Gln223Arg多态性(正常对照组104例,胃癌117例)。结果:胃癌组瘦素受体基因Gln223Arg的GG、GA和AA基因型表达频率分别为0.650、0.325和0.026,等位基因G和A频率分别为0.812和0.188;正常对照组基因型表达频率分别为0.798、0.192和0.010,G和A等位基因频率分别为0.894和0.106;Gln223Arg位点多态性的分布频率在对照组和病例组间差异有统计学意义(χ2=5.867,P<0.05)。结论:瘦素受体基因Gln223Arg多态性与汉族人群胃癌相关。
Objective: To understand the genotype distribution of leptin receptor Gln223Arg in Han population and to explore the relationship between the gene polymorphism and gastric cancer. Methods: Polymorphisms of leptin receptor gene Gln223Arg in 221 unrelated Chinese Han population were determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method (104 cases of normal control group, 117 cases of gastric cancer ). Results: The frequencies of GG, GA and AA genotypes of the leptin receptor gene Gln223Arg in gastric cancer group were 0.650,0.325 and 0.026, respectively. The frequencies of allele G and A were 0.812 and 0.188, respectively. The frequencies of genotypes in the normal control group were 0.798, 0.192 and 0.010 respectively. The frequencies of G and A alleles were 0.894 and 0.106, respectively. The distribution frequency of Gln223Arg polymorphism in control group and case group was statistically significant (χ2 = 5.867, P <0.05). Conclusion: Gln223Arg polymorphism of leptin receptor gene is associated with gastric cancer in Han population.
