目的利用中国毛虾制备具有ACE抑制活性及抗高血压活性的酶解产物。方法采用体外试验法检测中国毛虾5种常用蛋白酶酶解产物的ACE抑制活性,筛选出一种蛋白酶作为酶法制备中国毛虾ACE抑制产物用酶,并对其作用条件进行正交优化,利用原发性高血压大鼠评估酶解产物的抗高血压效应。结果胃蛋白酶优化后的酶解条件是:pH 2.4,温度41℃,酶解时间3 h,酶/底物比(E/S)3%,底物浓度8%。以该条件制备的酶解产物对ACE的抑制活性为IC500.65 mg/ml,以1.0 g/kg体重的剂量喂食原发性高血压大鼠4 h后收缩压降低3 886 Pa(29mmHg)。结论中国毛虾的胃蛋白酶酶解产物具有较强的ACE抑制活性及抗高血压活性。
Objective To prepare the enzymolysis product with ACE inhibitory activity and antihypertensive activity using Chinese shrimp. Methods The ACE inhibitory activities of 5 kinds of commonly used protease digested products were detected by in vitro assay. A protease was selected as an enzymatic method to prepare enzymes for inhibiting ACE inhibitors in Chinese shrimp, and its conditions were optimized orthogonally. Primary Hypertensive Rats Assess Antihypertensive Effects of Enzymatic Hydrolysates. Results The enzymolysis conditions optimized by pepsin were: pH 2.4, temperature 41°C, hydrolysis time 3 h, enzyme/substrate ratio (E/S) 3%, and substrate concentration 8%. The inhibitory activity of the enzymolyzed products prepared under these conditions on ACE was IC 500.65 mg/ml, and the systolic blood pressure was reduced by 3 886 Pa (29 mmHg) after feeding the rats with primary hypertension for 1.0 h at a dose of 1.0 g/kg body weight. Conclusion The pepsin hydrolysate of Chinese shrimp has strong ACE inhibitory activity and antihypertensive activity.
