目的:观察重组人卵泡刺激素(r FSH)对小鼠卵巢血管内皮生长因子(VEGF)以及整合素αvβ3表达的影响。方法:将4周龄C57BL/6J小鼠卵巢随机分为新鲜对照组(FCG)、玻璃化冻存对照组(VCG)、300 IU/L r FSH干预玻璃化冻存组(r FSH-VG)。通过对冻融卵巢内各级卵泡计数、免疫组织化学观察VEGF和整合素αvβ3在卵巢不同细胞中的定位,Western blotting检测VEGF、整合素αv和整合素β3蛋白表达量。结果:各级卵泡计数、VEGF及整合素β3的蛋白表达均为r FSHVG组优于VCG组(P<0.05),整合素αv的表达组间均无统计学差异(P>0.05)。结论:玻璃化冻融过程中添加r FSH可以上调VEGF及整合素β3蛋白的表达。
Objective: To observe the effect of recombinant human follicle stimulating hormone (r FSH) on the expression of vascular endothelial growth factor (VEGF) and integrin αvβ3 in mice. Methods: Four-week-old C57BL / 6J mice were randomly divided into three groups: fresh control group (FCG), vitrification control group (VCG) and 300 IU / L FSH intervention vitrification group (r FSH-VG). The levels of VEGF and integrin αvβ3 in different ovarian cells were observed by counting the number of follicles in the frozen-thawed ovary. The expression of VEGF, integrin αv and integrin β3 were detected by Western blotting. Results: The levels of follicle count, VEGF and integrin β3 were all significantly higher in FSHVG group than in VCG group (P <0.05). There was no significant difference in integrin αv between the two groups (P> 0.05). Conclusion: The addition of r FSH during vitrification can up-regulate the expression of VEGF and integrin β3.
