目的:探讨精子中精核蛋白含量与男性不育的关系、方法:从92例不育男性和16例正常生育男性的精子中提取精核蛋白.在醋酸-尿素聚丙烯酰胺凝胶(acid-ureaPAGE,pH4.7)中电泳.经薄层扫描,测得各蛋白区带(组蛋白和精核蛋白)的相对含量,分析并计算各蛋白区带的相对比值。结果:9例病人的精核蛋白完全缺乏。27例妻子流产男性精子畸形率升高明显(P<0.01)、HP1~3/TBP降低(P<0.01)。精子畸形率与精核蛋白的转化率成负相关。与正常生育组相比,92例男性不育症的总组蛋白(TH)与总碱性蛋白(TBP.包括TH、HP1、HP2、HP3)的比值增高;精核蛋白的含量由正常86.6±5.1%减少为64.1±14.4%(P<0.01);精子密度、精子活动力、畸形率、HP1/TBP、HP2+3/HP1等相差显著(P<0.01)。结论:精核蛋白含量异常的精子与畸形率、活动力、密度以及不明原因的流产等都具有相关性。
Objective: To investigate the relationship between the content of sperm nucleus and male infertility in sperm.Methods: The sperm nucleoprotein was extracted from sperm of 92 infertile men and 16 normal fertile men.After acid-urea polyacrylamide gel ureaPAGE, pH 4.7), and the relative content of each protein band (histone and nucleoprotein) was measured by TLC scanning. The relative ratio of each protein band was analyzed and calculated. RESULTS: Nine patients had complete absence of neucleoprotein. 27 cases of wives miscarriage male sperm deformity increased significantly (P <0.01), HP1 ~ 3 / TBP decreased (P <0.01). The sperm deformity rate is negatively correlated with the conversion rate of protamine. The ratio of total histone (TH) to total basic protein (TBP. Including TH, HP1, HP2, HP3) was increased in 92 male infertility patients compared with normal fertility group. The content of seminoprotein increased from normal 86.6 ± Sperm motility, malformation rate, HP1 / TBP, HP2 + 3 / HP1 and so on (P <0.01). CONCLUSIONS: Spermatozoa with abnormal nuclear protein content are associated with deformity, mobility, density, and unexplained abortion.
