为探讨人乙肝病毒(HHBV)DNA在树鼠句肝细胞内存在状态,对经血清学证明已被感染的树鼠句肝组织进行转移印迹杂交分析。结果,在实验感染早期的树鼠句肝细胞检出HBVDNA复制的中间体;在实验中晚期的动物肝细胞发现整合型的HBVDNA。表明树鼠句肝对HHBV敏感,可成为乙肝和肝细胞癌发病机理研究及治疗乙肝药物筛选的较好的实验动物模型。
In order to investigate the existence of human hepatitis B virus (HHBV) DNA in the rat hepatocytes, we analyzed the liver tissues of the rats that had been infected with serological evidence by western blot. As a result, HBVDNA-replicating intermediates were detected in rat hepatocytes at the early stages of experimental infection; and in the experiment, late-stage animal hepatocytes were found to have integrated HBVDNA. The results showed that the stump liver of rats was sensitive to HHBV and could be a good experimental animal model for studying the pathogenesis of hepatitis B and hepatocellular carcinoma and drug therapy of hepatitis B.
