采用RT_PCR差异显示法 ,从水稻 (OryzasativaL .)幼苗克隆了 1个受冷抑制表达的cDNA片段。该片段序列与水稻叶绿体基因组编码ATP合酶CF0 Ⅲ亚基的atpH基因完全同源 ,且覆盖了atpH基因编码区。以Northern杂交分析了水稻幼苗在冷处理不同时间后的atpH基因转录水平 ,结果表明 ,atpH基因的转录受冷抑制 ,在冷处理第 1天就明显下降 ,第 2天以后完全受抑制。
A cold-suppressed cDNA fragment was cloned from rice (Oryzasativa L.) seedlings using RT-PCR differential display. This fragment is completely homologous to the atpH gene encoding the ATP synthase CFO subunit of rice chloroplast genome and covers the coding region of atpH gene. The atpH gene transcription level of rice seedlings after cold treatment was analyzed by Northern blotting. The results showed that transcription of atpH gene was inhibited by cold, obviously decreased on the first day of cold treatment and completely inhibited after the second day.
