目的探讨乙型肝炎病毒(HBV)转染和Loxoribine刺激对Toll样受体7(TLR7)表达及干扰素-α(IFN-α)、白介素-12(IL-12)水平的影响。方法采用流式细胞术检测TLR7特异性配体Loxoribine刺激前后Hep G2和Hep G2.2.15细胞株TLR7的表达情况,ELISA法检测Loxoribine刺激培养的乙肝病毒携带组、慢性乙肝患者组和健康对照组外周血单个核细胞(PBMC)上清液中IFN-α、IL-12的水平。结果 Hep G2.2.15细胞内TLR7表达量较Hep G2细胞低(P<0.05),Loxoribine刺激培养后Hep G2和Hep G2.2.15细胞内TLR7的表达量均升高,但差异无统计学意义(P>0.05)。乙肝病毒携带组和慢性乙肝患者组PBMC上清液中IFN-α水平较健康对照组低(P<0.05),慢性乙肝患者组IL-12水平较乙肝病毒携带组和健康对照组高(P<0.05);Loxoribine刺激培养后健康对照组IFN-α、IL-12升高水平与乙肝病毒携带组和慢性乙肝患者组比较,差异有统计学意义(P<0.05);乙肝病毒携带组和慢性乙肝患者组比较,差异无统计学意义(P>0.05)。结论 HBV转染可下调TLR7的表达,乙肝病毒感染者外周血单个核细胞对Loxoribine刺激后TLR7介导的INF-α、IL-12分泌调节功能下降。
Objective To investigate the effect of hepatitis B virus (HBV) transfection and Loxoribine on the expression of Toll like receptor 7 (TLR7) and the levels of interferon-α (IFN-α) and interleukin-12 (IL- Methods Flow cytometry was used to detect the expression of TLR7 in Hep G2 and Hep G2.2.15 cell lines before and after stimulation with Loxoribine, a specific ligand of TLR7. The peripheral blood levels of Hep G2 and Hep G2.2.15 cell lines were detected by ELISA. Levels of IFN-α, IL-12 in supernatant of blood mononuclear cells (PBMCs). Results The expression of TLR7 in Hep G2.2.15 cells was lower than that in Hep G2 cells (P <0.05). The expression of TLR7 in Hep G2 and Hep G2.2.15 cells after Loxoribine stimulation was increased, but the difference was not statistically significant (P > 0.05). The levels of IFN-α in the PBMC supernatant of patients with hepatitis B virus infection and chronic hepatitis B patients were lower than those of the healthy controls (P <0.05), while the levels of IL-12 in patients with chronic hepatitis B were significantly higher than those of HBV carriers and healthy controls (P < 0.05). The elevated levels of IFN-α and IL-12 in healthy control group after Loxoribine stimulation were significantly different from those in HBV-infected and chronic hepatitis B patients (P <0.05); HBV-carrying group and chronic hepatitis B There was no significant difference between the two groups (P> 0.05). Conclusion HBV transfection can down-regulate the expression of TLR7. The monocytes from peripheral blood mononuclear cells infected with HBV can down-regulate TLR7-mediated secretion of IL-12 and INF-α after Loxoribine stimulation.
