目的 本文通过观察化疗药物诱导胰腺癌细胞凋亡过程中p5 3和bcl 2基因表达量的变化 ,探讨化疗药物诱导胰腺癌细胞发生凋亡过程与相关基因表达的关系。方法 采用斑点杂交及激光密度扫描技术对化疗药物配伍 :5 氟尿嘧啶 (5 FU)、丝裂霉素 (MMC)和表柔比星 (E ADM)诱导胰腺癌细胞株细胞发生凋亡过程中p5 3和bcl 2基因表达量的变化进行了检测分析。结果 化疗药物诱导胰腺癌细胞株细胞发生凋亡过程中 ,p5 3和bcl 2的基因表达量随细胞凋亡的发生分别升高和下降 ,与细胞凋亡数量的变化分别呈正相关和负相关。结论 化疗药物诱导胰腺癌细胞凋亡与p5 3和bcl 2基因表达相关
Objective To investigate the relationship between the expression of p53 and bcl 2 genes in pancreatic cancer cells induced by chemotherapeutic drugs and to explore the relationship between apoptosis and pancreatic cancer cell apoptosis. Methods Dot blot hybridization and laser densitometry were used for the combination of chemotherapeutic drugs: 5 fluorouracil (5 FU), mitomycin (MMC) and epirubicin (E ADM) induced apoptosis in pancreatic cancer cell lines. p5 3 Changes in the expression of bcl 2 gene and the detection were analyzed. Results During the apoptosis of pancreatic cancer cell lines induced by chemotherapeutic drugs, the expression levels of p53 and bcl-2 genes increased and decreased with the occurrence of apoptosis, respectively, and were positively and negatively correlated with the changes in the number of apoptosis. Conclusion Chemotherapy induces apoptosis of pancreatic cancer cells and is associated with p53 and bcl-2 gene expression
