目的:研究高三尖杉酯碱和血栓通对培养人翼状胬肉成纤维细胞周期性变化的影响。方法:对翼状胬肉组织中的成纤维细胞进行培养并传代为第3代~5代。用高三尖杉酯碱(10-5mol/L)及血栓通(1.5g/L)分别处理24h,对照组用生理盐水。应用PIDNA荧光染色及流式细胞仪检测和分析细胞周期的变化。结果:高三尖杉酯碱组和血栓通组G1群体细胞明显增多,S期细胞明显减少。与对照组比较差异均有明显意义(P<0.01),其中高三尖杉酯碱更为明显。结论:高三尖杉酯碱和血栓通对翼状胬肉成纤维细胞的增生均有明显抑制作用,以高三尖杉酯碱更显著。其作用机理可能与抑制细胞DNA合成有关。
Objective: To investigate the effects of homoharringtonine and xueshuantong on the periodontal changes of cultured human pterygium fibroblasts. Methods: Fibroblasts in pterygium were cultured and passaged for passage 3 to passage 5. The rats were treated with homoharringtonine (10-5mol / L) and Xueshuantong (1.5g / L) for 24 hours respectively. The control group was given saline. Application of PIDNA fluorescence staining and flow cytometry detection and analysis of cell cycle changes. Results: The cells in G1 homoharringtonine group and Xueshuantong group were significantly increased, and the cells in S phase were significantly decreased. Compared with the control group, the difference was significant (P <0.01), of which homoharringtonine was more obvious. Conclusion: Homoharringtonine and Xueshuantong can significantly inhibit the proliferation of pterygium fibroblasts, and homoharringtonine is more significant. Its mechanism may be related to inhibition of DNA synthesis.
